介绍
KRAS 突变在几种癌症中都有发现,例如结直肠癌、肺癌、甲状腺癌、胰腺癌、胆管癌。KRAS 突变通常出现在外显子 2 的密码子 12 和 13 上,导致 p21-ras 蛋白的生长信号异常变化。这种与细胞生长和分裂有关的异常变化可能通过传递过量信号引发癌症发展过程。KRAS 突变的存在通常与药物反应的预后标志相关。例如,KRAS 突变被认为是酪氨酸激酶抑制剂(如吉非替尼(Iressa)或厄洛替尼(Tarceva))药物反应的强预后标志。最近,KRAS 突变经常在结直肠癌中检测到,可能与用于结肠癌治疗的西妥昔单抗(Erbitux)或帕尼单抗(Vectibix)的药物反应有关。因此,检测 KRAS 突变对于确定结直肠癌或肺癌患者的药物耐药性,并有助于癌症治疗。
