什么是肽核酸(PNA)?
肽核酸 (Peptide Nucleic Acid,PNA) 是一种人工合成的类似于 DNA 或 RNA的聚合物,最初是由Peter E.Nielsen和他在哥本哈根大学的同事于1991年合成的 [1]。
肽核酸的基本单位是N-(2-氨基乙基)甘氨酸,由通过肽键连接形成聚合物 [2]。核酸碱基(ATCG)通过亚甲基羰基键而不是通常的脱氧核糖(DNA)或核糖(RNA)连接到主链上。与DNA或RNA双链相比,肽核酸与DNA或RNA可以形成更稳定的双链结构,并且这一结构代谢稳定。
特点
高结合亲和力
电荷中性骨架允许PNA通过Watson-Crick氢键与DNA或RNA结合,其结合亲和力明显高于带负电荷的DNA寡聚体 [3]。
高特异性
PNA序列对目标位点具有极高的特异性。如果有一个错配,则结合亲和力显著降低;如果有两个错配,则不会发生杂交 [4]。
高稳定性
PNA可以在DNA或者RNA降解的条件下依然表现出很强的化学稳定性和热稳定性 [5]。

由于PNA具有高结合亲和力,所以可以设计PNA靶向mRNA,通过空间干扰以影响其作用 [6]。
核酸检测
PNA独特的杂交特性和代谢稳定性使其非常适合于复杂的生物环境体系中的核酸检测 [7]。
基因编辑
PNA能够结合dsDNA并进行基因编辑。经过特定修饰以后,PNA介导的基因编辑的脱靶效应极弱 [8]。
